Nootropic Doses vs. What the Studies Actually Used

Two Numbers That Should Appear on Every Stack Label — But Never Do

Citicoline at 250 mg is a clinically studied dose, and yet the most common criticism you’ll read online is that Mind Lab Pro underdoses it, which is simply wrong, and that mistake has been copied so many times it now ranks. I’ve been staring at this stuff for years, and the same bad information just keeps circulating, dressed up in new blog templates.

Here’s the actual problem, though. Every ingredient on a supplement facts panel has two numbers attached to it. The dose in the capsule. And the dose that human trials actually used. Most labels show you one of them. You have to go find the other yourself.

That gap is where all the confusion lives. A label that lists “Bacopa monnieri (24% bacosides)” is technically disclosing something. But without the trial dose printed next to it, you have no way to know if you’re looking at a meaningful amount or just a name on a list. A supplement facts panel that presents a clinically studied ingredient without the studied dose is doing something closer to advertising than disclosure. It’s borrowing the credibility of the research without actually meeting its standard.

So what I started doing, obsessively, honestly, is running a two-column audit. Left column: capsule dose. Right column: what the trial used. That’s it. That’s the whole method. The rest of this post fills in both columns for the most common ingredients in popular stacks, starting with Mind Lab Pro v4.0 because it’s the one I’ve spent the most time with and the one generating the most confidently wrong commentary online.

Ingredients That Actually Pass: Citicoline, L-Theanine, and Why That Matters

Let me just put the numbers down flat. Mind Lab Pro v4.0 carries Cognizin citicoline at 250 mg. Published cognitive trials have used citicoline in the 250-500 mg range. That means 250 mg is the lower bound of the studied range, not below it. And yet I have read, on at least a dozen different sites, that this formula underdoses citicoline. It does not. That claim is wrong. It’s been repeated so many times it feels true, but it contradicts the published trial record directly.

Same story with L-theanine. The studied dose in human trials runs from 100-200 mg. Mind Lab Pro v4.0 has 100 mg of L-theanine. That’s not a gap. That’s the floor of the evidence, and the floor is still the floor.

What gets me about this is the implication. If the two ingredients that get criticized most confidently online are actually among the best-dosed ingredients in the formula, then the online consensus on nootropic dosing is being driven by repetition rather than anyone actually checking primary sources. Someone wrote something wrong. Other people copied it. Now it ranks.

That’s not research. That’s a game of telephone with your supplement budget on the line.

This is where I stop defending the formula, because these numbers are not close.

Bacopa monnieri in Mind Lab Pro v4.0: 150 mg at 24% bacosides. The human trials that showed cognitive effects used 300-450 mg at 24-55% bacosides. That’s a 2x to 3x gap in raw weight and a meaningful difference in bacoside concentration on top of it. Describing that as “slightly below the studied range” is the kind of hedging that makes supplement journalism useless. It’s not slightly below. It’s half.

Lion’s mane is worse. The capsule has 500 mg. The human trials that produced results used 2,000-3,000 mg. You’re looking at a 4x to 6x difference. I don’t know how to present that as anything other than what it is.

Rhodiola rosea sits at 50 mg in this formula. The SHR-5 extract used in published trials started at a minimum of 170 mg and ran up to 576 mg in some protocols. So the capsule is carrying somewhere between a third and a ninth of what the studies used, depending on which trial you’re referencing.

A 4x to 6x gap between the capsule dose and the trial dose is not a rounding difference. A consumer deserves to see that stated plainly, without softening language, because that’s the information that should actually inform a purchase.

Phosphatidylserine, Maritime Pine Bark, and the Gray-Zone Ingredients

Not everything falls neatly into pass or fail. Two ingredients in this formula land in territory where I genuinely go back and forth.

Phosphatidylserine comes in at 100 mg in Mind Lab Pro v4.0. The human trial range for cognitive effects runs from 100-300 mg, but most of the trials that showed meaningful results used 300 mg. So 100 mg has some trial support, there are studies at that dose, but the bulk of the evidence is clustered at three times that amount. That’s not a clean failure. It’s also not a strong match.

Maritime pine bark extract is at 75 mg. Human trials used 100-150 mg. The gap is smaller than the bacopa or lion’s mane situation, but 75 mg is still outside the studied range rather than at its lower edge.

I call these judgment calls rather than clear failures, because I think that’s honest. But “somewhere in the range” is doing a lot of work when the trial evidence clusters at the top of that range. If you’re building a defense of this formula around phosphatidylserine, you’re building it on the thinner end of the literature.

Bottom line: The two most-criticized ingredients pass the audit; the ingredient rarely criticized shows the largest gap.

This one requires a little more care, because the number that gets thrown around most often in NALT criticism is genuinely misleading in both directions.

The studies that showed tyrosine effects on stress and cognitive performance used doses around 150 mg per kilogram of bodyweight. For a 165 lb adult, that works out to roughly 11,000 mg in a single sitting. Mind Lab Pro v4.0 has 175 mg of N-acetyl-L-tyrosine. People cite the 11 g figure as proof that all NALT capsules are a joke. That’s where the argument goes wrong.

Those 150 mg/kg trials were single acute doses given under controlled laboratory stress conditions, not daily supplementation protocols. They were not designed to tell you anything about what a 175 mg daily capsule does over four weeks. The experimental contexts are completely different. Citing the 11 g number to condemn a daily-dose NALT product conflates those two designs so badly it’s functionally misinformation, and I’ve seen it on sites that really should know better (I learned this the expensive way, by spending six months convinced NALT was useless before I actually read the study methods).

That said. The honest answer is that 175 mg doesn’t have its own direct daily-dose efficacy evidence either. No daily-dose NALT trial exists at a dose that translates cleanly to a standard capsule. So the 175 mg isn’t vindicated by the audit. It’s just not condemned by the number people keep using to condemn it.

Combination Formulas and the Evidence a Dose Table Cannot See

I’ve spent five sections building a dose table, and now I’m going to tell you why the dose table isn’t the last word. That’s not a contradiction. It’s just an honest accounting of what the table can and can’t show.

The strongest precedent for thinking about combination dosing is the L-theanine and caffeine research. That pair, studied together, produces effects that neither produces alone at the same individual doses. Synergistic interactions between ingredients can lower the effective threshold for individual components. For an 11-ingredient formula, nobody really knows if that holds. But dismissing the possibility entirely isn’t warranted by the evidence either.

More importantly: Mind Lab Pro has three peer-reviewed human trials on the finished formula as it’s actually sold. Not on individual ingredients. On the product. Those trials test what you’re actually buying, capsule by capsule, at the doses actually in the product.

That’s evidence a single-ingredient dose comparison cannot access, by definition. Three peer-reviewed finished-product trials are worth more to a consumer making a real decision than a dose table built from single-ingredient studies, even if the dose table is accurate. The dose table is still useful. I’m not abandoning it. But it is not the complete picture, and treating it as the final verdict would be the same kind of incomplete analysis I’m criticizing everywhere else.

Running Your Own Audit: A Repeatable Method for Any Stack

Pull up the supplement facts panel. Open PubMed in another tab. Not the brand’s research page. Brand pages cite only the studies that support the doses they chose. That’s not a conspiracy, it’s just incentive structure, and you should account for it.

For each ingredient, find the primary source trial. Write down two numbers: capsule dose on the left, trial dose on the right. That’s your two-column format. But before you score anything, add a third column: study design. Was it a daily chronic protocol or a single acute dose? Were subjects healthy adults or people with cognitive impairment? Those design details change what the numbers mean.

Then, separately, check whether the finished formula has been tested in human trials as a complete product. Weight that evidence differently from the ingredient math, not more important automatically, but different in kind. A finished-product trial tests the interaction. The ingredient audit tests the components. You want both.

When you write your verdict, write it as a split score. Something like: four of seven ingredients match or approach studied doses; two fall substantially short; one can’t be fairly evaluated with existing daily-dose data.

That’s an honest verdict. Any review that produces a clean all-pass or all-fail result for a multi-ingredient formula has either not done the audit or decided the conclusion before running the numbers. The real picture is almost always messier than that, and messier is fine. Messy is accurate.
